Building a Scanning PALM Microscope
Development of System
The microscope system for this class was built using a Till Photonics iMic system as the main body of the confocal system, with some extensive modifications to the optical path. An overall optical path setup can be seen below:
Figure 1: A simple schematic of the optical path of the system. The Till iMic base provided housing for the objectives and a motorized stage to easily move the sample. The rest of the diagram is a custom optical path built by the students.
